CES buffered yeast extract (AYE) broth containing 10 g of N-(2-acetamido)-2-aminoethanesulfonic acid (ACES) buffer, 10 g of bovine serum albumin, 5 g of yeast extract, 0.4 g of l-cysteine-HCl, and 0.25 g of ferric pyrophosphate (all from Sigma, Deisenhofen, Germany) per liter of medium was used for in vitro cultivation of legionellae. All the ingredients were added to sterile distilled water, and the pH was adjusted with potassium hydroxide to 6.9. The AYE broth was filter sterilized and stored at 4°C.
